These Ladybugs were used in a care program enacted by Professor Fluttershy, presumably to teach the students of the School of Friendship about care and responsibility. Although very friendly, they triggered unpleasant memories in Twilight Sparkle, as when she was a kid, her house was swarmed with Ladybugs, who Shining Armor claimed were monsters with thousands of eyes, always watching her.
She never did get over her fear of ladybugs and instead gained a bizarre desire in seeing them float away, no doubt due to her botched Hypnosis session from Starlight Glimmer.
