Number One Assistant was the cute title Twilight Sparkle gave to Spike, her assistant.
The title eventually evolved into Friendship Ambassador, an official government title.
History
Spike took the title very seriously, as he took helping out Twilight and their friends very seriously. Though he reveled in the attention he got, he genuinely wanted to make Twilight and all of his other friends happy, and when he felt like he was replaced by Owlowiscious, he grew depressed and even vengeful towards the owl.
